Summary

Principles of cellular functions with regard to metabolism and energetics of biological systems, biocatalysis, breeding of industrial microorganisms, biotechnological processes and their implementation into engineering, isolation of system constituents. Implementation of biotechnologies into the system of technical science, application of biotechnologies in the preparation and processing of industrial mineral resources and waste water treatment. syllabus Lectures: 1. Introduction. Microbiology as a science. Classification of microorganisms. Classification of bacteria according to physiological properties. 2. A cell – composition of microbial cells. Bacteria - morphology, bacterial cell structure multiplication of bacteria classification of bacteria 3. Yeasts – cytology of yeasts multiplication of yeasts classification of yeasts 4. Moulds - basic characteristics classification of moulds Algae - basic characteristics classification of algae 5. Basic features of microorganisms’ metabolism Influence of external environment on microorganisms 6. Dynamics of growth and multiplication of microorganisms 7. Characteristics of bacteria of the Thiobacillus ferrooxidans genus 8. Mechanism of bacterial oxidation of sulphides by Thiobacillus ferrooxidans bacteria (direct and indirect). 9. Factors affecting the activities of T.F. bacteria Culture media for T.F. lO. Bacterial leaching method Technology of leaching ores. ll. Bacterial desulphurization of coal. l2. Přednášky: 1. Úvod. Mikrobiológia ako veda. Rozdelenie mikroorganizmov. Rozdelenie bakterií podľa fyziologických vlastností. 2. Bunka - zloženie mikrobiálnych buniek. Baktérie - morfológia, štruktúra bakteriálnej bunky rozmnožovanie baktérií rozdelenie baktérií 3. Kvasinky - cytológia kvasiniek rozmnožovanie kvasiniek rozdelenie kvasiniek 4. Pliesne - základné charakteristiky rozdelenie pliesní Riasy - základné charakteristiky rozdelenie rias 5. Základné rysy metabolizmu mikroorganizmov Vplyv vonkajšieho prostredia na mikroorganizmy 6. Dynamika rastu a množenia mikroorganizmov 7. Charakteristika baktérií rodu Thiobacillus ferrooxidans 8. Mechanizmus bakteriálnej oxidácie sulfidov baktériami Thiobacillus ferrooxidans (priama a nepriama). 9. Faktory ovplyvňujúce činnosť baktérií T.F. Kultivačné média pre T.F. lO.Technika postupu bakteriálneho lúhovania. Cvičení: l. Úvod. Základné podmienky pre prácu v mikrobiálnom laboratóriu. 2. Príprava živných pôd pre mikroorganizmy (všetky druhy pevných médií). Základné práce s mikrobiálnou kľučkou a s médiami. 3. Príprava mikrobiálnych preparátov. Grammove skúšky. 4. Identifikácia a sledovanie morfológie baktérií. 5. Identifikácia a sledovanie morfológie kvasiniek. 6. Stanovenie mikrobiálneho znečistenia vôd. 7. Príprava živných médií a množenie baktérií Thiobacillus ferrooxidans. 8. Príprava preparátov baktérií Thiobacillus ferrooxidans. 9. Počítanie baktérií - mikroskopická a MPN - metóda. lO.Odsírovanie hnedého uhlia baktériami T.F. ll.Aplikácia baktérii pri spracovaní odpadov z hutníctva a baníctva. l2.Vyhodnotenie testov z predchádzajúceho týždňa, stanovenie Fe2+ a počtu baktérií po týždni lúhovania. l3.Vyhodnotenie testov po 2.týždňoch lúhovania. l4.Vyhodnotenie testov po 3. týždňoch lúhovania a zhodnotenie aplikácie daného spôsobu úpravy. l5.Udelenie zápočtov.
