Subject aims expressed by acquired skills and competences
Target competences lie in gaining new knowledge and its application in observation, experiments and communication:
- to characterize the morphological characteristics of microorganisms, to understand phylogenetic relations of the different groups, to learn about and to term characteristic organisms of the individual taxons and to understand the significance of pathogenic as well as non-pathogenic species
- to apply the gained knowledge in dealing with practical tasks in seminars, to consider the consequences of a phenomenon from the environmental, sanitary as well as economic points of view; to build a positive attitude to nature
- to be able to work independently on the set tasks, to observe and objectively record the experiment results, to adhere to safe (aseptic) work principles when working with biological material and technical equipment
- to be able to work with expert/academic texts, to be familiar with the expert terminology, to be able to synthetize the obtained experimental data and to justify own approach in dealing with a biological problem

Summary
The subject is focused on consolidation and broadening of microbiological laboratory skills enabling isolation, cultivation and identification of microorganisms from waste and other environmental samples.
Learning outcomes of the subject are to deepen the knowledge and skills of students in working with microorganisms including the application of basic laboratory procedures, methods and techniques used in microbiological analyses of waste samples, including the identification of significant indicator groups of microorganisms and verification of the presence of significant pathogens.

Way of continuous check of knowledge in the course of semester
Practical skills are continuously checked when students work at tutorials.
The subject is concluded with a credit, students will submit reports from tutorials with elaborated tasks.
The credit is awarded on condition of 100% attendance at tutorials, elaborating and submitting reports, including completed assignments, and passing the credit test at 51 % at a minimum.

1. Principles of safe work in a microbiological laboratory
2. Equipment of a microbiological laboratory, basics of aseptic work
3. Preparation of nutrient media and their sterilization, conditions of cultivation of microorganisms
4. Bacteria and their morphological features on nutrient media
5. Microscopic filamentous fungi and their morphology on nutrient media
6. Yeast and yeast microorganisms, their morphological features
7. Cyanobacteria, microscopic algae and their morphology
8. Sampling and preparation of samples for microbiological analysis
9. Cultivation methods for determining the number of microorganisms in a sample
10. Microscopic methods for determining the number of microorganisms in a sample
11. Indirect methods for determining the number of microorganisms in a sample
12. Indicator microorganisms and identification of basic pathogens
13. Other methods of microbiological analysis of waste samples
