Subject aims expressed by acquired skills and competences

Obtaining basic knowledge not only about environmental sampling in relation to its share in the overall uncertainty of environmental analyzes but also in instrumental analytical methods. This will increase student readiness to solve analytic problems by selected instrumental methods. Obtaining the competence to work in an analytical laboratory with basic instrumental equipment, the ability to present and evaluate the results, to learn the correct principles of working with the sample.

Summary

The lecture provides information about selected modern instrumental methods of research and monitoring of the environment, its use and application. During the lectures, students will get acquainted with the principles of analytical methods that are commonly used in control and development analytical laboratories.

Way of continuous check of knowledge in the course of semester

Vědomosti studenta jsou ověřovány písemnou formou, přičemž je kladen důraz na porozumění danému tématu a aplikaci teoretických poznatků při řešení praktických úloh. Hodnocení zahrnuje i kolektivní hodnocení a sebehodnocení. Do celkového hodnocení je zahrnuta i aktivní práce studentů v hodině. Každá laboratoř začíná 5ti minutovým 6ti otázkovým testem, kterým student prokazuje, že četl návody a je schopen pracovat bezpečně a účelně.

Subject syllabus:

1. Definition of the issue. Information-gathering process. The analysis process. Evaluation of analytical results and practices, expression analysis results. Statistical results. Draft action arising from the data obtained. Principles of Good Laboratory Practice. Basics of chemometrics. 2. Sampling, the general principle and sources of the errors. Standard operating procedures for sampling; QA / QC system. Basic parameters sampling of abiotic and biotic matrices. 3. Treatment of the sample before analysis. Nondestructive methods. Destructive methods. Separation and concentration steps. 4. Qualitative analysis: classical system of dividing cations. Evidence heteroatoms in organic compounds. 5. Overview of the basic analytical methods and their general properties: methods based on weighing, measuring volume. The principles of individual methods; instrumentation; application in the analysis of environmental components; advantages/disadvantages. 6. An introduction to the instrumental methods. Use of both qualitative and quantitative. Definition of the issue. The analytical methods absolute and comparative. Rapid analytical methods - screening. Intended use. Advantages, disadvantages. Calibration methods. Trends in the instrumental methods of analysis. 7. Electrochemical methods based on the voltage measurement. Electrochemical methods based on the current measurement. The principles of individual methods; instrumentation; application in the analysis of environmental components; advantages/disadvantages. 8. Summary separation methods: Planar chromatography: TLC + HPTLC, PC, advantages/disadvantages. 9. Column chromatography: GC + HPLC. New trends: UHPLC, GC / MS, HPLC / MS. The principles of individual methods; instrumentation; application in the analysis of environmental components; advantages/disadvantages. 10. Electrophoresis. Mass spectrometry. The principles of individual methods; instrumentation; application in the analysis of environmental components; advantages/disadvantages. 11. Overview of the optical methods. Optical spectral device. Refractometry, interferometry, polarimetry and turbidimetry: Principles of the individual methods; instrumentation; application in the analysis of the environmental components; advantages/disadvantages. 12. Atomic emission spectral analysis. Flame photometry. Atomic absorption spectrometry. UV / Vis spectrometry. The principles of individual methods; instrumentation; application in the analysis of environmental components; advantages/disadvantages. 13. Luminescence analysis. Infrared spectrometry. Raman spectra; X-ray fluorescence spectrometry. Radiochemical methods. Understanding the various methods, instrumentation, application in the analysis of environmental components; advantages/disadvantages. 14. New trends in instrumental methods with applications in biotechnology.
