Obtaining fundamental knowledge in the field of molecular biology. Competences acquired: ability to define main concepts and to describe main approaches used in molecular and cell biology.

Summary
The course provides basic information about nucleic acids DNA and RNA, and introduction to basic mechanisms of molecular biology - replication, transcription and translation. Students will learn the basic methods of molecular biology used for the analysis of nucleic acids or proteins and their practical applications.

Subject syllabus:
Information macromolecules, structure of DNA and RNA. Molecular structure of genes and chromosomes. Replication of DNA, comparison of the replication in prokaryotes and eukaryotes. Errors and repairs of DNA. Transcription. Principles of the control of gene expression in prokaryotes and eukaryotes. Regulatory sequences, activators and repressors. Processing of eukaryotic mRNA. Translation. Structure of ribosomes. Genetic code and its importance. Structure of tRNA. Activation of amino acids. Initiation of the translation in prokaryotes and eukaryotes. Elongation. Termination. Posttranslational modifications of proteins. Chaperones and chaperonines. Fundamental methods of molecular biology. Isolation of DNA and RNA. Electrophoretic analysis of nucleic acids. Restriction analysis of DNA, construction of restriction maps, genome mapping. Polymerase chain reaction (PCR) in vitro. DNA sequencing, sequencing of genomes. Northern and Southern hybridization. Cloning strategies, transfer of DNA to bacterial and eukaryotic cells. Preparation of recombinant proteins. Site-directed mutagenesis. Transgenic organisms.
